Applications

Controlled bacterial cultivation; microbial physiology and metabolism studies; gene-expression studies; metabolic assays.

Formulation

Per liter: Na₂HPO₄·7H₂O 12.8 g; KH₂PO₄ 3 g; NaCl 0.5 g; NH₄Cl 1 g; MgSO₄ 2 mM; CaCl₂ 0.1 mM. MgSO₄ and CaCl₂ are filter sterilized and added separately. Carbon source and experiment-specific supplements are not included. Finished-product release pH: 7.2 ± 0.2.

Storage

Unopened: Store at 4–30°C. Ambient storage within this range is acceptable for shorter-term use; refrigeration is recommended for longer-term storage. After opening or supplementation: Refrigerate and use appropriate aseptic handling. Follow any more restrictive storage requirements associated with added supplements. Brief excursions above 30°C during normal shipping are acceptable; avoid prolonged excessive heat and temperatures low enough to cause freezing.

Shelf life

Unopened: 2 years. After opening: Up to 6 months when refrigerated and handled appropriately using aseptic technique. After supplementation, shelf life may be shorter and is limited by the stability and storage requirements of the added components.

Quality control

Growth tested with suitable microorganisms to confirm expected growth. After filling, bottles are observed for a minimum of 48 hours for evidence of unintended microbial growth as a contamination/sterility QC check. Finished-product pH, appearance, and fill volume are verified. Each finished unit receives a lot number for traceability. The base medium is sterilized by autoclaving; autoclave sterilization is monitored using biological/spore indicators and temperature-sensitive chemical indicator tape. MgSO₄ and CaCl₂ are filter sterilized and added separately.

M9 Minimal Broth is supplied as a sterile, pre-prepared, ready-to-supplement defined minimal-medium base for controlled bacterial cultivation and metabolic studies.

Add the desired carbon source and any experiment-specific supplements before inoculation, then use the completed medium under the conditions required by your organism and protocol.
